Super Mario World 2: Yoshi's Island is a platform video game acting as a prequel to 1990's Super Mario World. The game casts players as Yoshi as he escorts Baby Mario through 48 levels in order to reunite him with his brother Luigi, who had been kidnapped by Baby Bowser's minions. As a Super Mario series platformer, Yoshi runs and jumps to reach the end of the level while solving puzzles and collecting items. In a style new to the series, the game has a hand-drawn aesthetic and is the first to have Yoshi as its main character. The game introduces his signature abilities to flutter jump, produce eggs from swallowed enemies, and transform into vehicles.

Not everything is well in Dream Land. For some mysterious reason, the Dream Spring, a magical well that is the reservoir for all the dreams of the citizens of Dream Land, is no longer working. Using 20 unique tricks and your ability to steal your enemies' powers by swallowing them, you'll have to make your way through a horrific land filled with all kinds of nightmares. Recover the broken pieces of the Star Rod, and everyone in Dream Land will sleep peacefully once again. If you fail, the citizens of Dream Land will be subjected to a lifetime of terrible nightmares.

The game ditches the alternating two-player method in favor of simultaneous co-op. Another new feature is a password back-up. Both Mickey and Minnie are identical gameplay-wise, and the costumes they receive don't differ from each other in any aspect except aesthetically. The costumes include a sweeper, which can suck enemies and turn them into coins, a safari suit that can climb walls and swing from hooks and a cowboy suit that shoots corks and maneuvers by bouncing. The latter two outfits can be upgraded at a shop. The Genesis version of the game includes a cosmetic change for the stage 5 boss and an extra room in stage 3.

The game involves going through 8 stages (broken up into two parts each) of side-scrolling action. At the end of each stage is a boss monster that must be disposed of. Jerry moves by sliding across the ground and can latch on to walls and climb them. Jerry can also jump, stretch out to cling to ceilings, and also move at a faster pace. Jerry can consume balls that he finds during a stage and can then fire them out of his body at enemies blocking his path. Enemies can also be defeated by jumping on them and then flattening Jerry's body on them. Also, there are flowers that contain items for Jerry to collect. Items include life, which refills health, a 1-up that adds an extra life, and a seed, which will grow a climbable beanstalk when planted. If the player collects the scattered letters that appear in every stage that spell out the name Jerry, the player will earn two extra lives.
